目的 分析人胃癌组织中微小RNA-126(miR-126)及解整合素-金属蛋白酶9(ADAM9)的表达及其与临床病理参数的相关性,探讨二者与胃癌发生发展的关系。方法 收集2020年1月至2022年9月吉林省人民医院收治并经临床和病理组织学确诊为胃腺癌的46例患者手术切除的癌组织及距离癌组织3~5 cm的癌旁组织。采用荧光定量聚合酶链反应法检测胃癌组织和癌旁组织中miR-126及ADAM9 mRNA的表达,免疫组织化学法检测ADAM9蛋白的表达。记录患者年龄、性别、肿瘤大小、分化程度、肿瘤分期及转移等基本信息,分析miR-126及ADAM9的表达与胃癌患者临床病理参数的相关性。结果 胃癌组织中miR-126 mRNA的相对表达量显著低于癌旁组织[(0.53±0.24)比(1.00±0.00)](t=-8.613,P<0.001),ADAM9 mRNA的相对表达量显著高于癌旁组织[(1.43±0.56)比(1.00±0.00)](t=3.412,P=0.002)。ADAM9在胃癌组织中的阳性率显著高于癌旁组织[73.9%(34/46)比15.2%(7/46)],差异有统计学意义(χ2=29.723,P<0.001)。胃癌组织中miR-126的表达水平与患者肿瘤分化程度、TNM分期以及远处转移有关(均P<0.05)。胃癌组织中ADAM9的表达与患者肿瘤分化程度、淋巴结转移有关(均P<0.05)。结论 胃癌组织中miR-126的表达水平下调,ADAM9表达上调,二者之间可能存在负调节作用,且与胃癌的进展及转移关系密切。
Objective To analyze the expression of microRNA-126 (miR-126), a distegrinin and metalloprotease 9 (ADAM9) in gastric cancer and their correlation with clinicopathological parameters of patients, and to explore the relationship between them and the occurrence and development of gastric cancer. Methods A total of 46 patients with gastric adenocarcinoma admitted to Jilin Province People′s Hospital and diagnosed by clinical and histopathological examination from January 2020 to September 2022 were included in the study, and the resected cancer tissues and adjacent tissues about 3-5 cm away from the cancer were collected. The expressions of miR-126 and ADAM9 mRNA in gastric cancer tissues and adjacent tissues were detected by real-time quantitative polymerase chain reaction and the expression of ADAM9 protein was detected by immunohistochemistry. The basic information of patients such as age, gender, tumor size, differentiation degree, tumor stage and metastasis were recorded in detail. The correlation between the expression of miR-126 and ADAM9 and clinicopathological parameters of patients was analyzed. Results The relative expression of miR-126 mRNA in gastric cancer tissue was significantly lower than that in adjacent tissues[(0.53±0.24) vs (1.00±0.00)](t=-8.613, P<0.001), and the relative expression of ADAM9 mRNA was significantly higher than that in adjacent tissues[(1.43±0.56) vs (1.00±0.00)](t=3.412, P=0.002). The positive rate of ADAM9 in gastric cancer tissue was significantly higher than that in adjacent tissue cancer [73.9%(34/46) vs 15.2%(7/46)](χ2=29.723, P<0.001). The expression level of miR-126 in gastric cancer tissue was related to the degree of tumor differentiation, TNM staging, and distant metastasis of patients (all P<0.05). The expression of ADAM9 in gastric cancer tissue was related to the degree of tumor differentiation and lymph node metastasis in patients (both P<0.05). Conclusions The expression level of miR-126 is down-regulated and the expression of ADAM9 is up-regulated in gastric cancer tissues. There may be a negative regulatory effect between ADAM9 and miR-126, which is closely related to the progression and metastasis of gastric cancer.
