抗凋亡蛋白Livin和促凋亡蛋白Smac在绒毛组织中的表达及与稽留流产的关系

  • 目的    探讨抗凋亡蛋白Livin和促凋亡蛋白Smac在绒毛组织中的表达及与稽留流产的关系。方法    选取2015年1月至2016年1月在内蒙古医科大学附属医院妇产科确诊为稽留流产的患者30例为稽留流产组,同期选取自愿要求人工流产终止妊娠的早孕妇女30例作为正常早孕组。于人工流产术后取新鲜绒毛组织,通过链霉菌抗生物素蛋白-过氧化酶法检测Livin和Smac蛋白在2组绒毛组织中的表达情况,用JD801免疫组织化学图像分析仪检测阳性表达量。结果    稽留流产组Livin蛋白阳性平均吸光度值明显低于正常早孕组[(0.286±0.027)比(0.328±0.021)],差异有统计学意义(P<0.05)。稽留流产组Smac蛋白阳性平均吸光度值明显高于正常早孕组[(0.373±0.038)比(0.303±0.020)],差异有统计学意义(P<0.05)。Spearman检验结果显示,Livin与Smac蛋白在正常早孕组绒毛组织中的表达无相关性(r=-0.25,P>0.05),二者在稽留流产组绒毛组织中的表达呈明显负相关(r=-0.82,P<0.01)。结论    稽留流产绒毛组织中Livin低表达而Smac高表达,可能诱发绒毛细胞凋亡导致绒毛发育不良和退化,进而发生稽留流产。

  • Relation among expressions of anti-apoptotic protein Livin and pro-apoptotic protein Smac in chorionic villi with missed abortion

    【Abstract】Objective    To investigate the relation among expressions of anti-apoptotic protein Livin and pro-apoptotic protein Smac in chorionic villi with missed abortion. Methods    Thirty women with missed abortion(missed abortion group) and 30 women who required abortion(normal group) from January 2015 to January 2016 in the Affiliated Hospital of Inner Mongolia Medical University were enrolled. Chorion villi were sampled after abortion; expressions of Livin and Smac were detected by streptavidin-perosidase method and analyzed by the JD801 immunohistochemical image system. Results    The average absorbance of Livin positive expression in missed abortion group was significantly lower than that in normal group[(0.286±0.027) vs (0.328±0.021)](P<0.05). The average absorbance of Smac positive expression in missed abortion group was significantly higher than that in normal group[(0.373±0.038) vs (0.303±0.020)](P<0.05). Spearman analysis showed that expressions of Livin and Smac in chorion villi had no relation in normal group(r=-0.25, P>0.05); whereas there was a negative relation between expressions of Livin and Smac in chorion villi in missed abortion group(r=-0.82, P<0.01). Conclusion    Low expression of Livin and over expression of Smac may be associated with villous dysplasia and degeneration that can cause missed abortion.
