【摘要】目的 观察5-羟色胺2(5-HT2)受体激动剂及5-HT2受体拮抗剂对SD大鼠舌下神经核兴奋性的影响。方法 将16只成年雄性SD大鼠完全随机分为A、B组,各8只,均置入微透析导管于其舌下神经核。A组通过微透析导管向舌下神经核泵入新鲜人工脑脊液(ACSF)、新鲜5-HT2受体激动剂溶液,B组通过微透析导管向舌下神经核泵入新鲜ACSF、新鲜5-HT2受体拮抗剂溶液,监测A组未给药时、泵入ACSF时、泵入5-HT2受体激动剂溶液时和B组未给药时、泵入ACSF时、泵入5-HT2受体拮抗剂溶液时的颏舌肌肌电,利用区间测量方法计算颏舌肌肌电面积。结果 A组未给药时、泵入ACSF时、泵入5-HT2受体激动剂时大鼠颏舌肌肌电面积分别为(0.80±0.11)、(0.82±0.12)、(0.93±0.14)mV·s,未给药时与泵入ACSF时肌电面积比较,差异无统计学意义(P=0.280),泵入5-HT2受体激动剂时肌电面积明显大于未给药时与泵入ACSF时,差异均有统计学意义(P=0.005、0.005)。B组未给药时、泵入ACSF时、泵入5-HT2受体拮抗剂时大鼠颏舌肌肌电面积分别为(1.05±0.18)、(1.04±0.19)、(0.73±0.13)mV·s,未给药时与泵入ACSF时肌电面积比较,差异无统计学意义(P=0.955),泵入5-HT2受体拮抗剂时肌电面积明显小于未给药时与泵入ACSF时,差异均有统计学意义(P=0.003、0.003)。结论 5-HT2受体激动剂可以提高舌下神经核的兴奋性,5-HT2受体拮抗剂可以降低舌下神经核的兴奋性。
【Abstract】Objective To observe the effects of 5-hydroxytryptamine-2(5-HT2) receptor agonist and antagonist on hypoglossal nerve nuclear excitability in SD rats. Methods Sixteen adult male SD rats were randomly divided into group A and group B, with 8 rats in each group. The group A was injected fresh artificial cerebrospinal fluid(ACSF) and 5-HT2 receptor agonist, and the group B was injected ACSF and 5-HT2 receptor antagonist through microdialysis catheter into hypoglossal nucleus. Genioglossus electromyography was monitored before drug use, after giving ACSF and 5-HT2 agonist/antagonist. Results In the group A, genioglossus electromyography areas before drug use[(0.80±0.11)mV·s] and after giving ACSF[(0.82±0.12)mV·s] were similar(P=0.280); the electromyography area after giving 5-HT2 agonist[(0.93±0.14)mV·s] was significantly greater than that before drug use and after using ACSF(P=0.005, 0.005). In the group B, genioglossus electromyography areas before drug use[(1.05±0.18)mV·s] and after using ACSF[(1.04±0.19)mV·s] were similar(P=0.955); the electromyography area after giving 5-HT2 antagonist[(0.73±0.13)mV·s] was significantly smaller than that before drug use and after giving ACSF(P=0.003, 0.003). Conclusion 5-HT2 receptor agonist can improve the excitability of hypoglossal nucleus; 5-HT2 receptor antagonist can inhibit the excitability of hypoglossal nucleus.
