  • 目的 探讨西黄丸水提液抑制乳腺癌MDA-MB-231细胞迁移的分子机制。方法 通过细胞划痕实验分析不同浓度(57.515 g/L)西黄丸水提液对乳腺癌MDA-MB-231细胞迁移能力的影响。利用表皮生长因子(EGF)单独或联合加入西黄丸水提液,作用于乳腺癌MDA-MB-231细胞,通过蛋白质印迹法检测纤维连接蛋白(FN)、信号转导与转录激活因子3(STAT3)、磷酸化STAT3p-STAT3)、波形蛋白等蛋白的表达水平,分析西黄丸对于上皮细胞间质转化(EMT)的作用。结果 划痕实验显示,与对照组(西黄丸水提液浓度为0)相比,加入西黄丸水提液(57.515 g/L),36 h后肿瘤细胞的运动迁移能力受到抑制。与对照组(EGF浓度为0)相比,EGF浓度为406080100 μg/L时,FN相对表达量明显较高[(1.15±0.31)、(1.21±0.13)、(0.74±0.02)、(1.10±0.23)比(0.57±0.06)],EGF浓度为100 μg/L时,p-STAT3/STAT3明显较高[(1.60±0.08)比(0.96±0.15)](均P0.05)。使用60 μg/L EGF刺激乳腺癌MDA-MB-231细胞24 h后,加入不同浓度(057.515 g/L)西黄丸水提液作用24 h,结果显示,15 g/L的西黄丸水提液可降低FN与波形蛋白的表达水平。使用100 μg/L EGF刺激乳腺癌MDA-MB-231细胞24 h后,加入不同浓度(057.515 g/L)西黄丸水提液作用24 h,结果显示,15 g/L的西黄丸水提液可降低p-STAT3的表达水平。结论 西黄丸水提液可抑制乳腺癌MDA-MB-231细胞的迁移能力,同时可以在MDA-MB-231细胞中抑制因EGF刺激而增强的EMT过程。

  • Objective To investigate the molecular mechanism of Xihuang pill inhibiting the migration of breast cancer MDA-MB-231 cells. Methods The effects of different concentrations (5, 7.5, 15 g/L) of Xihuang pill on the migration ability of breast cancer MDA-MB-231 cells were analyzed by cell scratch test. Epidermal growth factor (EGF) alone or combined with Xihuang pill water extract were used to treat breast cancer MDA-MB-231 cells. Western blotting was used to detect the expression levels of fibronectin, signal transducer and activator of transcription 3 (STAT3), phosphorylated-STAT3 (p-STAT3) and vimentin, and the effect of Xihuang pill on epithelial-mesenchymal transition(EMT) was analyzed. Results  Scratch test showed that compared with the control group (the concentration of Xihuang pill was 0), the movement migration ability of tumor cells was inhibited after adding Xihuang pill water extract (5, 7.5, 15 g/L) for 36 h. The relative expressions of fibronectin were significantly higher at 40, 60, 80 and 100 μg/L EGF than those of the control group (EGF concentration was 0)(1.15±0.31), (1.21±0.13), (0.74±0.02), (1.10±0.23) vs (0.57±0.06), and p-STAT3/STAT3 was significantly higher at 100 μg/L EGF(1.60±0.08) vs (0.96±0.15)(all P0.05). The breast cancer MDA-MB-231 cells were stimulated with 60 μg/L EGF for 24 h, and then treated with different concentrations (0, 5, 7.5, 15 g/L) of Xihuang pill water extract for 24 h. It was showed that 15 g/L Xihuang pill water extract could reduce the expression levels of fibronectin and vimentin. The breast cancer MDA-MB-231 cells were stimulated with 100 μg/L EGF for 24 h, and then treated with different concentrations (0, 5, 7.5, 15 g/L) of Xihuang pill water extract for 24 h. It was showed that 15 g/L Xihuang pill water extract could reduce the expression level of p-STAT3. Conclusion Xihuang pill water extract can inhibit the migration ability of breast cancer MDA-MB-231 cells, and also can inhibit the EMT process enhanced by EGF stimulation in MDA-MB-231 cells.
