目的 探究姜黄素对缺血性脑卒中大鼠小胶质细胞增殖的影响。方法 选取40只健康SD大鼠为研究对象建立缺血性脑卒中模型,随机分为对照组与观察组,各20只。观察组予姜黄素0.5 mg/次、1次/d灌胃治疗7 d,对照组予相同容积0.9%氯化钠溶液灌胃。观察大鼠脑组织中小胶质细胞的数量及形态结构。采用Trizol法对Notch-1 mRNA表达情况进行测定。比较2组血浆炎症因子水平。结果 对照组大鼠小胶质细胞体积增大,细胞质内出现多个核体,大小不等,细胞突起增多并呈网状连接,细胞呈激活状体,染色细胞数目较多;观察组小胶质细胞数量相对较少,细胞相对结构形态较为正常,染色数目较少。观察组Notch-1 mRNA表达水平低于对照组[(0.365±0.072)比(0.933±0.022)],差异有统计学意义(t=0.572,P=0.015);观察组肿瘤坏死因子α、白细胞介素1β水平明显低于对照组[(380±21)ng/L比(873±27)ng/L、(289±20)ng/L比(901±35)ng/L],差异均有统计学意义(t=63.978、67.580,均P<0.001)。结论 姜黄素可在一定程度上抑制缺血性脑卒中大鼠小胶质细胞的增殖和激活,减轻炎症反应,其机制可能与Notch-1通路有关。
Objective To explore the effect of curcumin on proliferation of microglia in rats with ischemic stroke. Methods Totally 40 healthy SD rats were selected to establish the model of ischemic stroke. They were randomly divided into control group and observation group, with 20 rats in each group. The observation group was given curcumin oral administration 0.5 mg/time, once a day for 7 d, and the control group was given the same dose of 0.9% sodium chloride injection intragastric administration. The number and morphological structure of microglia in rats brain tissues were observed. The expression of Notch-1 mRNA was measured by Trizol method, and the levels of plasma inflammatory factors were compared between the two groups. Results In the control group, the volume of microglia increased; the number of nucleosomes appeared in the cytoplasm with different sizes; the number of cell protuberances increased and showed reticular connection. In the observation group, the number of microglia was relatively less, the cell structure and morphology were relatively normal, and the number of staining cells was less. The expression of Notch-1 mRNA in observation group was lower than that in control group [(0.365±0.072) vs (0.933±0.022)](t=0.572, P=0.015). The levels of tumor necrosis factor-α and interleukin-1β in observation group were lower than those in control group[(380±21)ng/L vs (873±27)ng/L,(289±20)ng/L vs (901±35)ng/L] (t=63.978, 67.580, both P<0.001). Conclusions Curcumin can inhibit the proliferation and activation of microglia in ischemic stroke rats to a certain extent, and reduce the inflammatory reaction. The mechanism may be related to Notch-1 pathway.
