  • 目的  初步探讨长链非编码RNA小核仁RNA宿主基因16lncRNA SNHG16)在宫颈癌组织中的表达及其与宫颈癌患者临床病理特征以及生存时间之间的相关性。方法 收集20141月至201512月在新疆医科大学附属肿瘤医院妇外科66例行手术切除并经病理学检查证实的宫颈癌患者的癌组织标本。同期收集本院20例子宫平滑肌瘤患者手术切除的正常子宫颈组织标本作为正常对照。采用实时荧光定量反转录聚合酶链反应技术检测宫颈癌组织、正常宫颈组织中lncRNA SNHG16的表达水平行组间比较;采用Kaplan-Meier法分析lncRNA SNHG16表达水平与宫颈癌患者年龄、病理特征及总体生存时间的关系。结果 宫颈癌组织中lncRNA SNHG16表达量明显高于正常宫颈组织[(3.01±0.29)比(0.99±0.08)],差异有统计学意义(P<0.05)。lncRNA SNHG16的表达量与宫颈癌患者的年龄、淋巴结转移情况没有显著相关性(均P>0.05),而与宫颈癌肿瘤直径、分化程度及肿瘤分期有显著相关性,肿瘤直径>4 cm者高于≤4 cm者、肿瘤低分化者高于中分化及高分化者、肿瘤分期Ⅱa2期者高于Ⅱa1期及Ⅰa~Ⅰb期者[(3.27±0.15)比(2.92±0.12),(3.29±0.14)比(2.99±0.11)、(2.94±0.20),(3.29±0.14)比(2.90±0.19)、(2.95±0.12)],差异均有统计学意义(均P<0.001)。生存分析结果 显示肿瘤组织中存在高表达lncRNA SNHG16的宫颈癌患者总生存时间明显短于低表达lncRNA SNHG16的宫颈癌患者(P<0.05)。结论 lncRNA SNHG16在宫颈癌组织高表达,其表达与宫颈癌肿瘤的直径、分化程度及肿瘤分期等临床病理特征以及生存时间显著相关。提示lncRNA SNHG16可能是宫颈癌致病分子,与宫颈癌发生发展相关。

  • Objective To investigate the expression of long non-coding RNA small nucleolar RNA host gene 16(lncRNA SNHG16) in cervical cancer tissues and its correlation with clinicopathological parameters and survival time in patients with cervical cancer. Methods Cancer tissue samples of 66 patients with cervical cancer who underwent surgical resection and confirmed by pathological examination in Department of Gynecological Surgery, Cancer Hospital Affiliated to Xinjiang Medical University from January 2014 to December 2015 were collected. At the same period, normal cervical tissue specimens from 20 patients with uterine leiomyoma were collected as normal controls. The expression levels of lncRNA SNHG16 in cervical cancer and normal cervical tissues were detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction, and were compared between the two groups. Kaplan-Meier method was used to analyze the correlation between the expression level of lncRNA SNHG16 and the age, pathological characteristics and overall survival time of patients with cervical cancer. Results The expression of lncRNA SNHG16 of cervical cancer tissues was significantly higher than that of normal cervical tissues(3.01±0.29) vs (0.99±0.08)(P<0.05). The expression of lncRNA SNHG16 was not significantly correlated with the age and lymph node metastasis of patients with cervical cancer (both P>0.05), but it was significantly correlated with the tumor diameter, degree of differentiation and tumor stage of cervical cancer, in which the tumor diameter >4 cm was higher than 4 cm, the tumor poorly differentiated was higher than that moderately and highly differentiated, and the tumor stage a2 was higher than stage a1 and a-b[(3.27±0.15 vs 2.92±0.12; 3.29±0.14 vs 2.99±0.11, 2.94±0.20;3.29±0.14 vs 2.90±0.19, 2.95±0.12)](all P<0.001). Survival analysis showed that the overall survival time of cervical cancer patients with high expression of lncRNA SNHG16 in tumor tissues was significantly shorter than that of cervical cancer patients with low expression of lncRNA SNHG16(P<0.05). Conclusions  LncRNA SNHG16 is highly expressed in cervical cancer tissues, and its expression is significantly correlated with clinicopathological characteristics such as the diameter, differentiation degree, tumor stage of cervical cancer and survival time of patients with cervical cancer. It is suggested that lncRNA SNHG16 may be a pathogenic molecule associated with the occurrence and development of cervical cancer.
