成纤维细胞生长因子2(FGF-2)是一种促进血管生成的因子,能够在多种细胞和组织中表达,并参与细胞迁移、细胞分化和各种发育过程。虽然FGF-2缺乏经典的分泌信号肽,但其可以通过结合高亲和力的跨膜FGF受体(FGFR)和辅助因子硫酸乙酰肝素蛋白多糖(HSPG)以FGF-2-HSPG-FGFR三元复合物的形式在细胞内发生磷酸化,并调节多种细胞的功能,参与正常伤口愈合和组织发育。近年来研究表明FGF-2对纤维化疾病的形成具有调节作用,在肺、肝、肾、心肌和皮肤纤维化中均存在FGF-2/FGFR信号通路的异常表达。本文对FGF-2在纤维化疾病中的研究进展进行综述,为FGF-2靶向治疗纤维化疾病提供参考。
Fibroblast growth factor-2(FGF-2), a member of angiogenic cytokines, can be expressed in a variety of cells and tissues and is involved in cell migration, cell differentiation and various developmental processes. Although FGF-2 lacks classical secretory signal peptide, it can bind to high affinity transmembrane FGF receptor (FGFR) and heparan sulfate proteoglycan (HSPG) and be phosphorylated in the form of FGF-2-HSPG-FGFR ternary complex, regulates the function of various cells and participates in the pathogenesis of normal wound healing and tissue development. In recent years, studies have shown that FGF-2 plays a regulatory role in the formation of fibrotic diseases. Abnormal expression of FGF-2/FGFR signaling pathway exists in lung, liver, kidney, heart and skin fibrosis. In this paper, the research progress of FGF-2 in fibrotic diseases is reviewed to provide a reference for the future research of FGF-2 targeted treatment in fibrotic diseases. font:minor-fareast;mso-hansi-font-family:Calibri; mso-hansi-theme-font:minor-latin'>倪晓容
