目的 研究宫颈癌组织中长链非编码RNA 染色质相关RNA 10(CAR10)和3-磷酸肌醇依赖性蛋白激酶1(PDPK1)的表达,分析二者对患者预后的评估价值。方法 选取2019年1月至2021年2月于新疆医科大学附属肿瘤医院接受治疗的136例宫颈癌患者。实时荧光定量聚合酶链反应检测宫颈癌组织CAR10、PDPK1 mRNA相对表达量。分析CAR10与PDPK1 mRNA的相关性。采用Log-rank检验比较不同CAR10、PDPK1 mRNA表达组宫颈癌患者生存曲线。采用Cox模型筛选宫颈癌患者预后的影响因素。结果 宫颈癌组织中CAR10、PDPK1 mRNA相对表达量均高于癌旁组织[(3.14±0.52)比(1.01±0.28)、(4.0±0.5)比(1.1±0.3)](均P<0.001)。宫颈癌组织CAR10与PDPK1 mRNA表达呈正相关(r=0.789,95%置信区间:0.347~0.897,P<0.001)。国际妇产科联盟(FIGO)分期ⅠB2~ⅡA2期宫颈癌组织CAR10和PDPK1 mRNA均高于ⅠA1~ⅠB1期患者(均P<0.001)。Log-rank检验结果显示,不同CAR10,PDPK1 mRNA表达组3年生存率比较,差异均有统计学意义(均P<0.05)。多因素Cox回归分析结果显示,FIGO分期ⅠB2~ⅡA2期、高CAR10和高PDPK1 mRNA均是影响宫颈癌患者预后的危险因素(均P<0.05)。结论 宫颈癌组织中CAR10和PDPK1 mRNA表达均显著上调,二者参与宫颈癌的进展过程,是评估患者预后的肿瘤标志物。
Objective To study the expression of long non-coding RNA chromatin associated RNA 10 (CAR10) and 3-phosphoinositol-dependent protein kinase 1(PDPK1) in cervical cancer tissues and analyze the evaluation value of both for the prognosis of patients. MethodA total of 136 patients with cervical cancer who were treated at Xinjiang Medical University Affiliated Cancer Hospital from January 2019 to February 2021 were selected. The relative expression levels of CAR10 and PDPK1 mRNA in cervical cancer tissues were detected by real-time fluorescence quantitative polymerase chain reaction. The correlation between CAR10 and PDPK1 mRNA was analyzed. Log-rank test was used to compare the survival curves of cervical cancer patients with different CAR10 and PDPK1 mRNA expression groups. Cox regression model was used to screen the prognostic factors of cervical cancer patients. Results The relative expression levels of CAR10 and PDPK1 mRNA in cervical cancer tissues were higher than those in adjacent tissues [(3.14±0.52) vs (1.01±0.28),(4.0±0.5) vs (1.1±0.3)](both P<0.001). There was a positive correlation between CAR10 and PDPK1 mRNA expression in cervical cancer (r=0.789, 95% confidence interval: 0.347-0.897, P<0.001). The mRNA levels of CAR10 and PDPK1 in international federation of gynecology and obstetrics (FIGO) stage ⅠB2-ⅡA2 cervical cancer were higher than those in stage ⅠA1-ⅠB1 patients(all P<0.001). Log-rank test showed that the 3-year survival rate of different CAR10 and PDPK1 mRNA expression groups was statistically significant (all P<0.05). Multivariate Cox regression analysis showed that FIGO stage ⅠB2-ⅡA2, high CAR10 mRNA and high PDPK1 mRNA were risk factors affecting the prognosis of cervical cancer patients (all P<0.05).Conclusion The expressions of CAR10 and PDPK1 mRNA are significantly up-regulated in cervical cancer tissues, which are involved in the progression of cervical cancer and are tumor markers for evaluating the prognosis of patients.
