  • 顺铂是许多癌症治疗的一线化疗药物,但因其肾脏毒性在临床应用中受到一些限制。近年研究发现,细胞衰老在顺铂所致急性肾损伤进程中发挥关键作用。细胞衰老表现为不可逆的细胞周期停滞,衰老细胞可以通过分泌多种炎性因子及促纤维化因子,构成衰老相关分泌表型,引起周围环境改变,进而驱动急性肾损伤向慢性肾脏病进展。因此靶向清除衰老细胞可能成为顺铂治疗期间肾脏保护的潜在治疗靶点,但其具体机制并不明确。本综述旨在阐述细胞衰老在顺铂诱导急性肾损伤及肾脏纤维化中的作用,期望能为防治顺铂毒副作用提供新的理论依据。

  • Cisplatin is a first-line chemotherapeutic agent for the treatment of various cancers, but its clinical application is limited due to its nephrotoxicity. Recent studies have found that cellular senescence plays a key role in the progression of cisplatin-induced acute kidney injury (AKI). Cellular senescence is characterized by irreversible cell cycle arrest, and senescent cells can form a senescence-associated secretory phenotype by secreting a variety of inflammatory and profibrotic factors, which alters the surrounding microenvironment and further drives the progression of AKI to chronic kidney disease. Therefore, targeted clearance of senescent cells may serve as a potential therapeutic target for renal protection during cisplatin treatment, while its specific mechanism remains unclear. This review aims to elaborate on the role of cellular senescence in cisplatin-induced AKI and renal fibrosis, hoping to provide a new theoretical basis for the prevention and treatment of cisplatin-induced toxic and side effects.
